Looking at the instructions for the shingles I will be buying, it states that one can use 4" or 5" or 6" spacing/exposure. Since this is a complete tear off job, and these will be the first layer, does it make any difference which exposure I use? I have a 6/12 pitch, on 30# felt. My guess is 5", but I don't know.

On a 6/12 pitch the 4 or 5" is fine. I never recommend the 6" although the manufacturer allows it. 5" exposure should put your tabs of top shingle even with the top of the rain slot on the bottom shingle. To me this is best installation with 6/12 using best stretch of your shingle yet best insurance not to leak.
If you roof pitch was 2/12 to 5/12 pitch I would recommend the 4" being less coverage of the material but best ensurance to repel rain.
In my opinion your thought of 5" exposure on 6/12 is fine ensuring the meltdown hold to work best utilizing the most safe use of your material.
